I would read the guidelines Nextdoor has posted for members. I pretty much details the capability of this website. However, They do not have this capability or it would be built into the message so that you select notification of a response.

Send a request to the recipient asking if they had an opportunity to read your message and that a yes or no response is adequate.

I doubt NEXTDOOR will send you a delivery or "opened" confirmation as this is typically done by email apps, not Website postings.
